Project Description:
Development of cell-seeded TE valves using decellularised valvular scaffolds (ESR6)

The aim of this Fellowship is to explore the feasibility of developing a living valve replacement using decellularised human or porcine semilunar valves, reseeded with human cells and physically conditioned in a bioreactor. Aortic and pulmonary valves will be decellularised according to previously established protocols, and will be functionalised with a cohort of different bioactive peptides to enhance cell colonisation. The functionalised scaffolds will be assessed against their untreated counterparts in terms of structural/biomechanical alterations, surface morphology, function and durability in collaboration with other ESR projects within the TECAS-ITN. The functionalised scaffolds will be seeded statically with either human stromal cells, or bone marrow stem cells to optimise cell/scaffold-type combination and seeding conditions. Optimally-seeded scaffolds will be conditioned in a pulsatile bioreactor under different conditions to optimise valve regeneration. Statically-seeded and conditioned valves will be assessed in terms of cell phenotype, structural/biomechanical integrity and neo-matrix synthesis to assess the effect of dynamic conditioning on valve regeneration.
