Project Description: Investigation of the growth and calcification potential of fibrin gel-based TE valves in vivo (ESR8)

The aim of this Fellowship is to investigate the remodelling, growth and calcification potential of valves fabricated from fibrin gel and seeded with smooth muscle cells (SMCs) and endothelial cells (ECs), in the growing ovine model. Ovine fibrin valves will be fabricated according to a previously developed method, and assessed in terms of their durability and calcification potential in other ESR projects within the TECAS-ITN. Half of the fabricated valves will be treated with an anticalcification treatment that will be developed in a separate ESR project within TECAS-ITN and the rest will be left untreated. Autologous treated and untreated valves will be seeded statically with autologous SMCs and ECs, which will be isolated from the umbilical cords of the lambs that will be harvested during birth. The seeded valves will be cultivated in a pulsatile bioreactor for 21 days, and subsequently, implanted in the pulmonary position of 2-3 months old lambs. Treated and untreated valves will be explanted after 4, 8, 12 and 30 months, and assessed in terms of cell phenotype, extracellular matrix remodelling, valve growth and localisation of calcification.
