Project Description: Evaluation of non-destructive online markers for tissue development (ESR3)

The aim of this Fellowship is to develop a non-destructive method for assessing the quality of TE constructs during tissue development and maturity. Specifically, this project will investigate the amount and course of split-off products of extracellular matrix (ECM) proteins in the culture media and correlate it to their amount, location and distribution in the TE construct, and the resulting construct biomechanics. Initially, the project will focus on a model frame-fixed ovine-fibrin scaffold seeded with heart valve interstitial cells (VICs) or mesenchymal stem cells (MSCs) in static cultures. The ECM protein content of the constructs will be assessed by histology and biochemistry, and the construct integrity by biomechanical testing. The results will be correlated to the amount of ECM proteins in the culture media of the constructs. The method will be validated with bioreactorconditioned cell-seeded fibrin valves, decellularised aortic and percutaneous valves, and myocardial patches developed in other projects within the TECAS-ITN.
