Project Description: In vitro calcification models for cardiovascular scaffolds (ESR4)

The aim of this Fellowship is to develop static and dynamic in vitro models for assessing the calcification potential of cardiovascular scaffolds. The Fellowship will investigate decellularised porcine pericardial valves and aortic/pulmonary valve roots, decellularised porcine/bovine pericardial patches, and ovine-fibrin valvular scaffolds, to investigate the modes and occurrence of calcification under static and simulated physiological and non-physiological dynamic conditions. The project will also investigate the effectiveness of anticalcification treatments, established in another project within the TECAS-ITN, on inducing anticalcification resistance to the scaffolds. The work will focus on establishing the calcifying solutions and utilising them in static and dynamic cultures in established functional simulation bioreactors. The decellularised valvular scaffolds and patches will be prepared according to previously established protocols, whereas the fibrin scaffolds will be contributed by another ESR project within the TECAS-ITN. Following the static and dynamic cultures, the scaffolds will be tested histologically, biomechanically, and by spectroscopy and chemical analysis, to assess the mode, occurrence and effect of calcification.
