Project Description: Experimental investigation of ?-actin remodelling (ESR2)

  

The aim of this Fellowship is to develop an experimental system, which will be able to induce uniaxial or biaxial dynamic loading on cell-seeded scaffolds, with a view to measuring cell-induced traction forces. The setup will be designed to enable visualisation and inspection of cellular properties, such as ?-actin and viability, and collagen orientation, using staining in a 3D setting. The work will focus on the development of collagen- and fibrin-based micro-tissue model systems, employing micro-fabrication technology and heart valve interstitial cells, which will be integrated in a cyclic stretching device under aseptic conditions. Subsequently, ?-actin and collagen fibre orientation and distribution will be visualised and recorded under cyclic strain using confocal microscopy, and the cellular-level traction forces generated will be quantified using cantilever deflection measurements. The ?-actin and collagen fibre architecture will be compared against histological sections of full-scale bioreactor-conditioned tissues and, together with the measured cellular-level forces, in the validation of computational models developed in other projects within the TECAS-ITN.
