Project Description:
Assessment of the durability of valvular scaffolds for clinical use (ESR13)

The aim of this Fellowship is to assess the durability of a cohort of valvular scaffolds, including decellularised porcine aortic/pulmonary valves, decellularised porcine/bovine pericardium percutaneous valves (PHVs) and ovine fibrin valves, under real-time and accelerated functional testing. Pulmonary and aortic valves will be decellularised according to previously-developed protocols, whereas PHVs and fibrin valves will be contributed by other ESR projects within the TECAS-ITN. The scaffolds will be subjected to sterilisation and anticalcification treatments developed in other ESR projects within the TECAS-ITN. Treated and untreated scaffolds, together with native-fresh aortic and pulmonary valves (control), and commercial bioprosthetic valves (control), will be subjected to cyclic fatigue testing under pulsatile flow and aseptic conditions. The testing will initially be conducted under different frequencies, pressures, flows and test fluids to establish the optimal testing methodology. The durability of the treated scaffolds will be assessed in terms of structural/biomechanical alterations and hydrodynamic function, and will be compared against the durability of their untreated counterparts, and the durability of the native-fresh and bioprosthetic valves.
