Project Description: Anticalcification treatments for TE scaffolds (ESR5)

The aim of this Fellowship is to evaluate the effectiveness and optimise the performance of a range of current and novel treatments on inducing anticalcific protection to decellularised porcine pericardial valves and aortic/pulmonary valve roots, decellularised porcine/bovine pericardial patches, and ovine-fibrin valvular scaffolds. The decellularised valvular scaffolds and patches will be prepared according to previously established protocols, whereas the fibrin scaffolds will be contributed by another ESR project within the TECAS-ITN. Once prepared, the scaffolds will be treated with a cohort of anticalcification treatments, and will be assessed against their untreated counterparts in terms of induced cytotoxicity, structural/biomechanical alterations, surface morphology and durability. The effectiveness of the treatments on inducing anticalcific resistance to the scaffolds will be assessed in static and dynamic in vitro calcification model systems, which will be established in another ESR project within the TECAS-ITN.
